  • Descrição: Atherosclerosis is a chronic inflammatory disease responsible for more than 40% of the deaths in industrial countries. The presence of mLDL and autoantibodies against mLDL are implicated as important predictors of disease severity. Our aims were to separate the components of human mLDL and used the separated components as target for autoantibodies in ELISA meassuring IgG autoantibodies. Human LDL was oxidized in vitro (20 uM CuSO4, 18h). The mLDL, was separated on a Superdex-200 column (Amersham-Pharmacia-Biosciences) and monitored with a diode-array detector (Shimadzu). The lipid content was determined by enzimatic methods (LABTEST) and Fiske-Subbarow for determination of phospholipids. We obtained 4 peaks detected at 280nm and one major lipid containing peak. The autoantibody profile against lipid-free peak and adjacent were analyzed using sera from normal healthy individuals (n=30) and individuals with coronary artery (CAD) (n=35). Three fractions containing high mass components exhibited high reactivity with sera from healthy individuals as compared with those of CAD individuals. Those containing low mass components exhibited the inverse property. A number of putative antigens on mLDL are recognized by autoantibodies. In here we show that the reactivity of these are not only against antigens of lidpid-derived nature but against fragments from the protein moiety of LDL, apoB. In addition and most interstingly using these fragments we disclosed a disease-related pattern
