  • Descrição: Aims: implantation of the blastocyst is made possible through changes in uterine tissues induced by ovarian hormones followed with different responses of the different cell populations. although many of the changes have been described, attribution to cellular mechanisms remained evasive due to difficulties to localize hormones at the cellular level. micro-radioautography was used to identify cellular targets of estradiol during implantation and decidual transformation. Methods: Mice were mated and on days 4, 5, 6, and 7 of pregnancy injected i.v. each with 0.2ug of 3H-estradiol (S.A. 140 C/mM). One hour after injection, tissues were excised, freeze-mounted, frozen section cut at 4um, and thaw-mounted on emulsion-coated slides, exposed for several months, then developed and stained. Results: At all time points, strongest nuclear receptor binding is present in cells of the basal stroma and predecidua, glandular epithelium, vascular pericytes, and arteriolar muscle cells. ) Nuclear labeling decreases in maturing decidual cells and in lumenal epithelium in the vicinity of the implant. Small high density estradiol receptor cells, some closely associated with capillaries, are observed among large polyploid decidual cells. Topographic changes of estradiol binding exist within the endometrium and between antimesometrial and mesometrial sites. Inner and outer muscularis show at all times intermediate nuclear labeling, while migratory blood cells appear unlabeled.
    Conclusions: The results indicate complex and extensive actions of estradiol on specific cell populations related to differences in receptor binding, time, dose and topography, pertaining to genomic effects on proliferation, differentiation, secretion and others
